ABSTRACT

ObjectiveTo analyze the quality of life and related factors in patients with depression, and to provide evidence for formulating the corresponding measures to improve the patients' quality of life. MethodsA total of 117 inpatients who met the International Classification of Diseases, tenth edition(ICD-10) for depression were selected in Central People's Hospital of Zhanjiang, Affiliated Hospital of Guangdong Medical University and Guangdong 999 Brain Hospital from November 19, 2018 to September 7, 2019. The values of patients' clinical objective indicators consisted of routine blood test, routine urine examination, blood biochemical tests and blood gas analysis were collected by reviewing their medical records, and all subjects were assessed using Quality of Life Instruments for Chronic Diseases-Depression V2.0 [QLICD-DE(V2.0)]. Simple correlation analysis was used to explore the correlation between the QLICD-DE (V2.0) scores and various clinical objective indicators, and multiple linear regression was adopted to further screen factors affecting quality of life of patients with depression. ResultsSimple correlation analysis suggested that QLICD-DE (V2.0) total score was positively correlated with total protein and platelet distribution width in patients with depression (r=0.198, 0.281, P<0.05 or 0.01), and negatively correlated with hematocrit (r=-0.300, P<0.01). Multiple linear regression analysis showed that albumin-to-globulin ratio and treatment compliance (B=-19.836, -3.711, P<0.05 or 0.01) were influential factors for physical function. Platelet distribution width (B=2.706, P<0.01) was an influential factor for psychological function. Bilirubin (B=-6.375, P<0.05) was an influential factor for social function. Platelet distribution width (B=2.101, P<0.05) was an influencing factor for specific module. The albumin-to-globulin ratio and platelet distribution width (B=-10.653, 2.114, P<0.05 or 0.01) were influencing factors for total score of QLICD-DE (V2.0). ConclusionTreatment compliance and clinical objective indicators platelet distribution width, bilirubin and albumin-to-globulin ratio may be the influencing factors of quality of life in patients with depression.

ABSTRACT

Objective:To construct lentiviral vector which can overexpression miR-137 and produce lentivirus by lentivirus packaging system, and to explore its infection efficiency and expression in HEK293T cells. Methods: miR-137 sequence was chemically synthesized and cloned into lentiviral vector GV209, and the recombinant plasmid containing human miR-137 was obtained and identified. Then miR-137 recombinant plasmid together with two helper plasmids were transfected into HEK293T cells using Lipofectamine 2000.After the HEK293T cells were infected in multiplicity of infection(MOI) 40 for 48 h, the expression of green fluorescent protein (GFP) was observed by fluorescence microscope and the expression level of miR-137 was detected by fluorescence quantitative PCR.Results:The sequencing results showed that the inserted gene sequence was completely consistent with the published human miR-137 gene sequence in GenBank. The GFP was observed in the HEK293T cells infected with miR-137 overexpression lentivirus under fluorescence microscope. The fluorescence quantitative PCR results showed that the expression level of miR-137 in the cells infected with overexpression lentivirus was 12.74 times higher than that in the control cells. Conclusion:The lentivirus containing miR-137 gene is successful packaged, and it could efficiently infect the HEK293T cells.

ABSTRACT

Objective To investigate the aassociation of a microRNA-137 (miR-137) polymorphism, single nucleotide polymorphism (SNP) rs1625579, with neurocognitive function in patients with schizophrenia. Methods A total of 250 patients with schizophrenia were included in this study. The positive and negative syndrome scale (PANSS) was used to evaluate patients. The brief assessment of cognition in schizophrenia (BACS) scale was used to determine neurocongnitive functions in patients. Blood samples of patients were collected, and SNaPshot technique was used to compare the neurocognitive functions of different genotypes of rs1625579. Results The genotypes of TT, GT and GG were 221 (88.4%), 28 (11.2%) and 1(0.4%). There was no significant difference in PANSS score between TT genotype carriers and G allele (GG+GT) carriers. The detection of BACS showed that the digit sequencing score was significantly lower in patients with TT genotype than that of G allele (GG+GT) carrier ( P<0.05). There were no significant differences in other scores of BACS evaluation between two groups of patients. Conclusion The miR-137 polymorphism influences the working memory performance of schizophrenic patients in Chinese Han population.

ABSTRACT

BACKGROUND:At present, most general hospitals in China treat patients with neurosis and mental diseases merely by means of clinical psychological consultation, which cannot meet the requirements of social service for metal disorders. Is it feasible to establish open psychology wards?OBJECTIVE:To investigate the feasibility of establishing open psychology ward in general hospitals for patients with neurosis and mental diseases.DESIGN:Case analysis based on patients.SETTING:Department of Psychology of Guangdong Medical College Affiliated Hospital.PARTICIPANTS:Totally 900 patients with various types of neurosis were admitted to the Department of Psychology of Guangdong Medical College Affiliated Hospital between July 2000 and March 2003.There were 688 patients with different kinds of psychoneurosis and112 cases of mental diseases. Of them, there were 523 men and 377women aged from 11 to 80 years.with the mean age of (42±8) years and hospitalization duration of(17.0±6.5) days.METHODS:Open management consisting of open ward, supporting system, humanized restriction, collective physical exercise and psychological support was adopted.Symptom checklist (SCL-90) was used by trained psychological doctors to assess the therapeutic effects in the 900 patients with neurosis and mental diseases after two-month rehabilitation.Meanwhile,169 patients with depression were examined with Hamilton rating scale for depression(HAMD) at 2,4,8 weeks of treatment.patients.RESULTS:Open psychology ward could provide an environment for patients to live and make free contact with the outside world. Of the 900patients with various neurosis and metal diseases,531 (59.0%) had recovery to normal,264(29.4%) had obvious improvement,81(9.0%) had improvement, and 24 (2.6%) had no therapeutic effect, with the total effective rate of 97.4% .None of the patients had any accident. HAMD score of the 169 patients with depression was (32.3±3.6) before treatment, was obviously lower at 2,4 and 8 weeks of treatment [(17.4±4.2),(11.5±4.8) and(5.8±3.6),respectively](P < 0.01).CONCLUSION:The therapeutic effect of the open psychology ward in the general hospital on various neuropathies and mental diseases was 97.4% in the 900 cases, with no accident occurring during hospitalization.Depression of the 169 patients was obviously improved, indicating that it isfeasible to establish open psychology ward in general hospitals.
